The longitudinal SR are thinner projects, that run between the terminal cisternae/junctional SR, and are the location where ion channels necessary for calcium ion absorption are most abundant. This is the primary site of calcium release. T-tubules are closely associated with a specific region of the SR, known as the terminal cisternae in skeletal muscle, with a distance of roughly 12 nanometers, separating them. Cardiac and skeletal muscle cells contain structures called transverse tubules (T-tubules), which are extensions of the cell membrane that travel into the centre of the cell. The sarcoplasmic reticulum is a network of tubules that extend throughout muscle cells, wrapping around (but not in direct contact with) the myofibrils (contractile units of the cell). Therefore, it is vital that calcium ion levels are controlled tightly, and can be released into the cell when necessary and then removed from the cell. This means that too much calcium within the cells can lead to hardening ( calcification) of certain intracellular structures, including the mitochondria, leading to cell death. Calcium is used to make calcium carbonate (found in chalk) and calcium phosphate, two compounds that the body uses to make teeth and bones. This means that small increases in calcium ions within the cell are easily detected and can bring about important cellular changes (the calcium is said to be a second messenger). Calcium ion levels are kept relatively constant, with the concentration of calcium ions within a cell being 10,000 times smaller than the concentration of calcium ions outside the cell. The main function of the SR is to store calcium ions (Ca 2+). The sarcoplasmic reticulum ( SR) is a membrane-bound structure found within muscle cells that is similar to the smooth endoplasmic reticulum in other cells. The thinner projections, running horizontally between two terminal cisternae are the longitudinal sections of the SR. Therefore, redox and calcium homeostasis and protein glycosylation in the ER provide novel drug-targets for medical treatment in a wide array of diseases.Menbrane-bound structure in muscle cells for storing calcium A cartoon section of skeletal muscle, showing T-tubules running deep into the centre of the cell between two terminal cisternae/junctional SR. The unique set of enzymes, selection of metabolites, and characteristic ion and redox milieu of the luminal compartment strongly argue against the general permeability of the ER membrane.Īlterations in the luminal environment can trigger the unfolded protein response, a common event in a variety of pathological conditions. Mounting evidence supports the existence of a real barrier between the ER lumen and the cytosol. Ca(2+)-dependence of certain ER chaperones is a subject of intensive research. Transport of the required intermediates across the ER membrane and maintenance of the luminal redox conditions and Ca(2+) ion concentration are indispensable for appropriate protein maturation.Ĭooperation of enzymes and transporters to maintain a thiol-oxidizing milieu in the ER lumen has been recently elucidated. Compartmentation plays a crucial role in the post-translational modifications, such as oxidative folding and N-glycosylation in the ER lumen. Proteins destined to secretion and exposure on the cell surface are synthesized and processed in the extracellular-like environment of the endoplasmic reticulum (ER) of higher eukaryotic cells.

If you’re working on a personal project, SketchUp Make is for you. We know there’s been some confusion about choosing the right version of SketchUp in the past, so we want to be absolutely clear about this going forward. Read all about it here: re(Introducing) SketchUp Make. It’s a reflection of our commitment to our “3D for Everyone” mantra, and I’m super proud to say it’s here to stay. The latter is still free, international, and aimed squarely at every treehouse builder, 3D printing wizard, and pinewood derby all-star in the universe. Now the word “SketchUp” refers to a product family of which there are two members: SketchUp Pro and SketchUp Make. We decided that the free version of SketchUp needed a name and a brand of its own. SketchUp Make: A new brand for an old favorite We’ve written another one just for that purpose: SketchUp Pro 2013: A closer look at LayOut. The complete list of details about what’s new in SketchUp Pro 2013 is too long to include in this blog post. In this version, we’ve added hatching and other pattern fills, speedier vector rendering, better zoom, more useful callouts and other improvements we think you’ll love. For that, we’re continuing to turn LayOut in SketchUp Pro into a full-fledged tool for creating scaled, annotated and dimensioned drawings from your models. We’ve heard that you’d like to take your SketchUp models farther into the documentation part of your workflow. LayOut in SketchUp Pro 2013: More tools for turning your models into drawings No more wondering why this can’t all be easier. No more unpacking files and folders into precise locations in your file system. No more digging around for your plugins directory. Clicking a plugin’s “Install” button takes care of just about everything that used to make Ruby scripts so cumbersome to use. When you visit the Extension Warehouse, you’ll notice it’s every bit a modern app store: most-popular lists, user reviews, download statistics, introductory videos and more. And best of all, it all happens right inside SketchUp.īy far the best way to get to the Extension Warehouse is by clicking its icon in SketchUp 2013’s main toolbar. Using the Extension Warehouse to find, install and update plugins is a simple operation. This one new feature is actually dozens (eventually hundreds) of new features, all ready and waiting for you to discover. No more.įor 2013, we built a repository of extensions-an Extension Warehouse, in our parlance-that provides a one-stop shop for anyone looking to customize their copy of SketchUp. Historically, these extensions have been crazy useful, laughably affordable, and (for the most part) incredibly difficult to find. Over the years, our beloved plugin developers-the folks who use our Ruby scripting tools to build add-ons for SketchUp-have created some truly amazing features. We think you’ll be really excited about both-we certainly are.Įxtension Warehouse: A smarter approach to SketchUp pluginsĬan SketchUp do X, Y, or Z? Extension Warehouse is the place to find out. For this release, we focused on two things: building an ecosystem that makes it easier for millions of SketchUp modelers to find and use the plugins and extensions they need, and continuing to turbocharge SketchUp Pro’s documentation and presentation features (in the form of LayOut). Less than a year after joining the Trimble family, it’s our pleasure to announce the arrival of SketchUp 2013. Making the most of its gleefully shallow setting in Beverly Hills, Heckerling never shortchanges the intellect or innate goodness of her less than deep protagonist, a foresight that ultimately helps to define it from similar films of its ilk. Carried deftly by the preternaturally charming Alicia Silverstone in a star making role as the immaculate and perpetually optimistic Cher and flanked by similarly shiny-haired co-stars in Stacey Dash, Brittany Murphy (RIP!) and the baby-faced Paul Rudd, the film is a classic high school comedy of genius proportions. Easy to like and simply stuffed with charm, Clueless is a breezy comedy with some knowing bite thanks to sharp-toothed scripting from writer/director Amy Heckerling and such a pure-hearted center that it’s as impossible to dislike as its bubbly protagonist. Let’s face it: Clueless is the Jeff Goldblum of ‘90s movies. – Dave TrumboreĬast: Alicia Silverstone, Stacey Dash, Paul Rudd, and Brittany Murphy That’s a lesson that’s every bit as uplifting today as it was 30 years ago. It’s through his quest to reunite with his family that he actually manages to bring about meaningful change in mouse society at large in the New World. Rather than be stopped in his tracks by those who would choose to prey on him when he gets lost in the big city, Fievel makes a variety of friends from all classes, nationalities, and backgrounds throughout his travels. The underdog in this case is actually a tiny, charming mouse of Russian Jewish heritage by the name of Fievel Mousekewitz. You would be forgiven for thinking that this animated tale that starts with an anti-Semitic attack, a troubled ocean crossing, and the separation of a family of America-bound immigrants is not exactly “feel good.” But it’s in the first act of this under-appreciated classic that the dismal stakes are established so that the film’s ultimate conclusion is that much more rewarding. No-one’s certain whether they’ll be needed or not. That’ll be crucial considering - much like uncertainties around possible unreliability - teams can’t rely on experience alone when plotting a route to the flag thanks to the all-new car.īrake wear as emerged as a potential early curveball, with each car allowed two front brake pad changes in Sandown, although pad changes won’t be mandatory until Bathurst. Sandown offers an opportunity for them to stretch their legs before the Great Race. With the grid reducing by one car in 2024, there’s extra pressure to nail the opportunity.Ī sprinkling of longer format races, like Newcastle and Townsville, have kept the strategists sharp, but Bathurst has otherwise stood alone as a real test of team tactics. Meanwhile, there’s always the prospect of some co-drivers being on audition for a full-time drive next season. The pair delivered Walkinshaw Andretti United its first Bathurst triumph in a decade, and with Holdsworth fresh off a full-time season in 2022, they’re tipped to pick up where they left off. The reunion of 2021 Bathurst winners Chaz Mostert and Lee Holdsworth will be another one to watch. He’s also a Sandown 500 winner with Warren Luff in 2016.Īll attention will be on Tander’s first ever outing with the Blue Oval, and partnered with a fellow Bathurst winner in Reynolds, it’s a combination not to be underestimated. Since stepping away from the full-time seat at the end of 2018 he’d won three endurance events from seven entries, including a pair of Bathurst victories with Shane van Gisbergen in the last three seasons. It’s a new alignment for 2023 after Tander sensationally defected from Triple Eight and General Motors to the Ford-backed Grove squad. While there are plenty of good stories throughout the grid’s set of combinations, let’s start with David Reynolds and Garth Tander. Success requires a different mindset - one that acknowledges compromise is the only way to maximise the overall result. In recent months throttle body sizes have been tweaked, which has aided driveability for the Mustang, but the shift-cut delta when changing gears has reportedly been widened in recognition of Ford’s power delivery being underwhelming through the rev range. Hand in hand with reliability are the ongoing rumblings about parity between the Mustang and the Camaro.ĭespite Ford’s apparent fightback in Tailem Bend, where Mustang drivers claimed six of nine podium finishes over three races - albeit behind the dominant Camaro of Brodie Kostecki - concerns have remained that the two models haven’t been equalised ahead of the crucial enduros double-header.Ĭhanges were made to the Mustang after a parity inquest was opened earlier in the year following a dire run of results for Ford, and the aerodynamically demanding Bend circuit seemed to positively validate them.īut straight-line speed is still an issue for some Ford team bosses.ĭrag was thought to be playing a role, but focus has more recently returned to engines and transmission. WILL THE PARITY DEBATE REAR ITS HEAD AGAIN? Those teams that have best understood their packages, that can best finesse them to go the distance, will have an oversized advantage relative to the rest of the season. Reliability is always a factor in endurance racing - it’s right in the name. They’ll now be doubling that, and in surprisingly warm conditions for Melbourne too to really test what they’re made of. Most of these problems have been understood and covered.īut the most any car has had to race up to this stage of the season is 250 kilometres. None of this is necessarily outside the scope of the sorts of problems you’d expect from such an overhaul of the technical regulations, particularly given the car’s gestation during the pandemic. There are even concerns that the Gen3 car is less easily repairable than previous models, though undoubtedly this has and will continue to improve as mechanics become more familiar with their machines. There have been those spectacular fires that have torched a few Mustangs as well as chronic problems with steering racks, gear towers, cooling, control parts - all sorts. Unsurprisingly for a clean-sheet design, the Gen3 machine hasn’t had a season of completely smooth sailing. While there’s a broader question about how the all-new cars will behave over the race distance - bearing in mind they’ll be up for double duty in Bathurst - more pertinent is whether they’re mechanically up to the task of going the distance. css file with that and run NW.JS, but no luck, the ratio and other stuff changed, so seems can be changed but no idea of what i'm doing, really.
